This sequence change replaces arginine, which is basic and polar, with serine, which is neutral and polar, at codon 299 of the TYR protein (p.Arg299Ser). This variant is present in population databases (rs61754374, gnomAD 0.03%). This missense change has been observed in individual(s) with ocular albinism (PMID: 13680365, 16570240, 18701257, 31077556). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 99587). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt TYR protein function with a positive predictive value of 80%. This variant disrupts the p.Arg299 amino acid residue in TYR. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 1642278, 28112372, 28266639). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic. -

The TYR c.895C>A variant is predicted to result in the amino acid substitution p.Arg299Ser. This variant has been reported in the compound heterozygous state in several individuals with oculocutaneous albinism (see for examples King et al. 2003. PubMed ID: 13680365; Zhong et al. 2019. PubMed ID: 31077556). Alternate substitutions of this amino acid (p.Arg299His and p.Arg299Cys) have also been reported in individuals with oculocutaneous albinism (Zhong et al. 2019. PubMed ID: 31077556). This c.895C>A (p.Arg299Ser) variant is reported in 0.038% of alleles in individuals of East Asian descent in gnomAD (http://gnomad.broadinstitute.org/variant/11-88924445-C-A), and has been classified as pathogenic by multiple independent submitters to the ClinVar database (https://www.ncbi.nlm.nih.gov/clinvar/variation/99587/). Given the evidence, we interpret c.895C>A (p.Arg299Ser) as pathogenic. -
