The c.939dupA pathogenic mutation, located in coding exon 11 of the MLH1 gene, results from a duplication of A at nucleotide position 939, causing a translational frameshift with a predicted alternate stop codon (p.V314Sfs*48). This mutation, designated as "938 A ins" was detected in an individual with MSI-H colorectal cancer at age 34 (Samowitz WS et al. Gastroenterology, 2001 Oct;121:830-8). In addition to the clinical data presented in the literature, this variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
