This sequence change replaces lysine, which is basic and polar, with asparagine, which is neutral and polar, at codon 1336 of the ABCC8 protein (p.Lys1336Asn). This variant is present in population databases (rs67767715, gnomAD 0.007%). This missense change has been observed in individual(s) with diffuse congenital hyperinsulinism and neonatal diabetes mellitus (PMID: 15562009, 32041611). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is also known as Lys1337Asn. ClinVar contains an entry for this variant (Variation ID: 551087). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt ABCC8 protein function with a negative predictive value of 95%. Experimental studies have shown that this missense change does not substantially affect ABCC8 function (PMID: 17575084). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
