The proline at codon 1112 is replaced by histidine, an amino acid with similar properties. This alteration has been reported as a secondary cardiac variant in an exome cohort and was detected in a cardiomyopathy/arrhythmia genetic testing cohort; however, clinical details are limited (Ng D et al. Circ Cardiovasc Genet, 2013 Aug;6:337-46; van Lint FHM et al. Neth Heart J, 2019 Jun;27:304-309). This alteration has also been reported in sudden death and arrhythmogenic right ventricular cardiomyopathy (ARVC) cohorts (Hertz CL et al. Int J Legal Med, 2016 Jan;130:91-102; Medeiros-Domingo A et al. Europace, 2017 Jun;19:1063-1069). The variant was also seen in cases of cardiomyopathy and was seen with variants in other cardiac-related genes (Refaat MM et al. BMC Med Genomics, 2019 Feb;12:33; Mastroianno S et al. Ann Noninvasive Electrocardiol, 2020 May;25:e12687; Paz-Cruz E et al. Cardiol Res, 2023 Oct;14:409-415). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
