Variant summary: GLB1 c.152T>C (p.Ile51Thr) results in a non-conservative amino acid change located in the Glycoside hydrolase 35, catalytic domain (IPR031330) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 249530 control chromosomes. c.152T>C has been reported in the literature in multiple individuals affected with Mucopolysaccharidosis Type IVB (Morquio Syndrome B) (example, Yoshida_1991). These data indicate that the variant is very likely to be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function, however, does not allow convincing conclusions about the variant effect (Iwasaki_2006). The following publications have been ascertained in the context of this evaluation (PMID: 16617000, 1907800). ClinVar contains an entry for this variant (Variation ID: 926). Based on the evidence outlined above, the variant was classified as pathogenic. -
