This variant is predicted to introduce a premature translational stop signal in exon 31 of COL1A1 and is expected to lead to degradation of the affected transcript. Loss-of-function variants in COL1A1 are an established cause of osteogenesis imperfecta (PMID: 27509835). This variant is very rare in the Genome Aggregation Database (v2.1.1). We have observed this variant in the Shriners Hospital for Children Canada variant database in an unrelated individual with a diagnosis of osteogenesis imperfecta. -
This sequence change creates a premature translational stop signal (p.Arg697*) in the COL1A1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in COL1A1 are known to be pathogenic (PMID: 7942841, 9295084, 9443882). This variant is present in population databases (rs72651642, gnomAD 0.007%). This premature translational stop signal has been observed in individuals with osteogenesis imperfecta (PMID: 8808594, 21667357, 28378289).
