The LDB3 gene has multiple clinically relevant transcripts. This variant occurs in alternate transcript NM_007078.3, and corresponds to NM_001080116.1:c.*18702T>A in the primary transcript. This sequence change replaces tyrosine, which is neutral and polar, with asparagine, which is neutral and polar, at codon 597 of the LDB3 protein (p.Tyr597Asn). This variant is present in population databases (rs727503131, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with LDB3-related conditions. Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The p.Y597N variant (also known as c.1789T>A), located in coding exon 10 of the LDB3 gene, results from a T to A substitution at nucleotide position 1789. The tyrosine at codon 597 is replaced by asparagine, an amino acid with dissimilar properties. This variant was detected in a cardiomyopathy/arrhythmia genetic testing cohort; however, clinical details were limited, and additional cardiac variants were detected in some cases (van Lint FHM et al. Neth Heart J, 2019 Jun;27:304-309). This variant has also been reported in an unexplained sudden cardiac arrest cohort (Grondin S et al. Eur Heart J, 2022 Aug;43:3071-3081). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
