Genes affected
MYH9 (HGNC:7579): (myosin heavy chain 9) This gene encodes a conventional non-muscle myosin; this protein should not be confused with the unconventional myosin-9a or 9b (MYO9A or MYO9B). The encoded protein is a myosin IIA heavy chain that contains an IQ domain and a myosin head-like domain which is involved in several important functions, including cytokinesis, cell motility and maintenance of cell shape. Defects in this gene have been associated with non-syndromic sensorineural deafness autosomal dominant type 17, Epstein syndrome, Alport syndrome with macrothrombocytopenia, Sebastian syndrome, Fechtner syndrome and macrothrombocytopenia with progressive sensorineural deafness. The Arg1936Trp vari ant in MYH9 has not been reported in the literature or in large population studi es. The arginine (Arg) residue at position 1936 is not conserved across species , with sloth having a Trp. Although additional information is needed to fully as sess the clinical significance of this variant; the lack of conservation suggest s a more likely benign role. -

Uncertain significance, criteria provided, single submitterclinical testingJohns Hopkins Genomics, Johns Hopkins UniversityApr 04, 2022This MYH9 missense variant (rs727503281) is rare (<0.1%) in a large population dataset (gnomAD: 8/250532 total alleles; 0.003193%; no homozygotes). It has an entry in ClinVar (Variation ID 164412), but has not ben reported in the literature, to our knowledge. Three bioinformatic tools queried predict that this substitution would be deleterious, and the arginine residue at this position is evolutionarily conserved across all species assessed. We consider the clinical significance of c.5806C>T to be uncertain at this time. -

Uncertain significance, no assertion criteria providedclinical testingPreventionGenetics, part of Exact SciencesJun 03, 2024The MYH9 c.5806C>T variant is predicted to result in the amino acid substitution p.Arg1936Trp. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.0098% of alleles in individuals of South Asian descent in gnomAD. At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
