MYBPC3 is expressed exclusively in heart muscle and is a key regulator of cardiac contraction. Mutations in this gene are a frequent cause of familial hypertrophic cardiomyopathy. This changes the amino acid from a glutamine to a stop codon within coding exon 20. This alteration has previously been described in a hypertrophic cardiomyopathy (HCM) cohort (Page SP et al. Circ Cardiovasc Genet. 2012;5:156-66). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
