Genes affected
DIAPH1 (HGNC:2876): (diaphanous related formin 1) This gene is a homolog of the Drosophila diaphanous gene, and has been linked to autosomal dominant, fully penetrant, nonsyndromic sensorineural progressive low-frequency hearing loss. Actin polymerization involves proteins known to interact with diaphanous protein in Drosophila and mouse. It has therefore been speculated that this gene may have a role in the regulation of actin polymerization in hair cells of the inner ear. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

ACMG classification

Classification made for transcript

Verdict is Benign. Variant got -14 ACMG points.

BP4
Computational evidence support a benign effect (BayesDel_noAF=-0.72).
BP6
Variant 5-141526375-C-T is Benign according to our data. Variant chr5-141526375-C-T is described in ClinVar as [Likely_benign]. Clinvar id is 179152.Status of the report is criteria_provided_multiple_submitters_no_conflicts, 2 stars. Variant chr5-141526375-C-T is described in Lovd as [Likely_benign].
BP7
Synonymous conserved (PhyloP=-2.72 with no splicing effect.
BS2
High AC in GnomAdExome4 at 13 AD gene. Variant has AC lower than other variant known as pathogenic in the gene, so the strength is limited to Supporting.
