Genes affected
JUP (HGNC:6207): (junction plakoglobin) This gene encodes a major cytoplasmic protein which is the only known constituent common to submembranous plaques of both desmosomes and intermediate junctions. This protein forms distinct complexes with cadherins and desmosomal cadherins and is a member of the catenin family since it contains a distinct repeating amino acid motif called the armadillo repeat. Mutation in this gene has been associated with Naxos disease. Alternative splicing occurs in this gene; however, not all transcripts have been fully described. [provided by RefSeq, Jul 2008]

Uncertain significance, criteria provided, single submitter | clinical testing | Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine | Aug 16, 2013 | The Ala566Thr variant in JUP has not been reported in individuals with cardiomyo pathy or in large population studies. The affected amino acid is conserved in ev olution, suggesting that a change would not be tolerated. Other computational an alyses (biochemical amino acid properties, AlignGVGD, PolyPhen2, and SIFT) do no t provide strong support for or against an impact to the protein. Additional inf ormation is needed to fully assess the clinical significance of the Ala566Thr va riant. - |

Uncertain significance, criteria provided, single submitter | clinical testing | GeneDx | Feb 28, 2023 | Has not been previously published as pathogenic or benign to our knowledge; Not observed at significant frequency in large population cohorts (gnomAD); In silico analysis supports that this missense variant does not alter protein structure/function - |
Cardiovascular phenotype Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Oct 09, 2024 | The p.A566T variant (also known as c.1696G>A), located in coding exon 9 of the JUP gene, results from a G to A substitution at nucleotide position 1696. The alanine at codon 566 is replaced by threonine, an amino acid with similar properties. This variant has been reported in an individual who underwent genetic testing for arrhythmogenic cardiomyopathy, but clinical details were limited (Murphy J et al. Ir J Med Sci, 2024 Aug;193:1775-1785). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. - |
