This protein forms distinct complexes with cadherins and desmosomal cadherins and is a member of the catenin family since it contains a distinct repeating amino acid motif called the armadillo repeat. The alanine at codon 566 is replaced by threonine, an amino acid with similar properties. This variant has been reported in an individual who underwent genetic testing for arrhythmogenic cardiomyopathy, but clinical details were limited (Murphy J et al. Ir J Med Sci, 2024 Aug;193:1775-1785). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
