This variant is associated with the following publications: (PMID: 31983221) -
not specified Uncertain:1
The Ile31985Thr variant in TTN has not been previously reported in individuals w ith cardiomyopathy and was absent from large population studies. Isoleucine (Ile ) at position 31985 is not conserved in evolution and 1 mammal (chimp) carries a threonine (Thr) at this position, raising the possibility that this change may be tolerated. Additional computational prediction tools suggest that this varian t may not impact the protein, though this information is not predictive enough t o rule out pathogenicity. In summary, the clinical significance of the Ile31985T hr variant is uncertain. -
Autosomal recessive limb-girdle muscular dystrophy type 2J;C1858763:Dilated cardiomyopathy 1G Uncertain:1
This sequence change replaces isoleucine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 34553 of the TTN protein (p.Ile34553Thr). This variant is present in population databases (rs727505196, gnomAD 0.01%). This missense change has been observed in individual(s) with dilated cardiomyopathy (PMID: 31983221; internal data). ClinVar contains an entry for this variant (Variation ID: 179887). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Not Available". The threonine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. This variant is located in the M band of TTN (PMID: 25589632). Non-truncating variants in this region may be relevant for neuromuscular disorders, but have not been definitively shown to cause cardiomyopathy (PMID: 23975875). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
Cardiovascular phenotype Uncertain:1
The p.I25488T variant (also known as c.76463T>C), located in coding exon 185 of the TTN gene, results from a T to C substitution at nucleotide position 76463. The isoleucine at codon 25488 is replaced by threonine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
