The Val211Ile in MAP2K1 has not previously been reported in individuals with cli nical features of a Noonan spectrum disorder or in large population studies. The Val residue at this position is highly conserved across evolutionarily distant species, though computational prediction tools do not provide strong support for or against an impact to the protein. In summary, the clinical significance of t he Val211Ile variant is uncertain. -

This sequence change replaces valine with isoleucine at codon 211 of the MAP2K1 protein (p.Val211Ile). The valine residue is highly conserved and there is a small physicochemical difference between valine and isoleucine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with a MAP2K1-related disease. ClinVar contains an entry for this variant (Variation ID: 179898). Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
