The p.K500E variant (also known as c.1498A>G), located in coding exon 4 of the TMPO gene, results from an A to G substitution at nucleotide position 1498. The lysine at codon 500 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
The Lys500Glu variant in TMPO has not been previously reported in individuals wi th cardiomyopathy or in large population studies. Computational prediction tools and conservation analysis do not provide strong support for or against an impac t to the protein.
