The c.4961-3delC variant in LAMA4 has not been previously reported in individual s with cardiomyopathy, but has been identified in 1/8254 of European American ch romosomes by the NHLBI Exome Sequencing Project (http://evs.gs.washington.edu/EV S/). This variant is located in the 3' splice region and computational tools sug gest a possible impact to splicing. However, this information is not predictive enough to determine pathogenicity. In summary, the clinical significance of the c.4961-3delC variant is uncertain. -
Dilated cardiomyopathy 1JJ Uncertain:1
This sequence change falls in intron 35 of the LAMA4 gene. It does not directly change the encoded amino acid sequence of the LAMA4 protein. It affects a nucleotide within the consensus splice site. This variant is present in population databases (rs727505356, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with LAMA4-related conditions. ClinVar contains an entry for this variant (Variation ID: 180112). Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
Cardiovascular phenotype Uncertain:1
The c.4961-3delC intronic variant, located in intron 34 of the LAMA4 gene, results from a deletion of one nucleotide within intron 34 of the LAMA4 gene. This nucleotide position is well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
