This frequency is not significantly higher than estimated for a pathogenic variant in DSP causing Arrhythmogenic Right Ventricular Dysplasia/Cardiomyopathy (4.4e-05 vs 0.0002), allowing no conclusion about variant significance. The c.5167_5169delGAA variant (also known as p.E1723del) is located in coding exon 23 of the DSP gene. This variant results from an in-frame GAA deletion at nucleotide positions 5167 to 5169. This results in the in-frame deletion of a glutamic acid at codon 1723. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear. -
