Hypertrophic cardiomyopathy Uncertain:1
This sequence change replaces glycine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 1260 of the MYBPC3 protein (p.Gly1260Asp). This variant is present in population databases (rs730880606, gnomAD 0.1%). This missense change has been observed in individual(s) with dilated cardiomyopathy (PMID: 20215591). This missense change has been observed to co-occur in individuals with a different variant in MYBPC3 that has been determined to be pathogenic (PMID: 20215591; internal data), but the significance of this finding is unclear. ClinVar contains an entry for this variant (Variation ID: 181025). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The c.3779G>A (p.G1260D) alteration is located in exon 33 (coding exon 33) of the MYBPC3 gene. This alteration results from a G to A substitution at nucleotide position 3779, causing the glycine (G) at amino acid position 1260 to be replaced by an aspartic acid (D). Based on data from gnomAD, the A allele has an overall frequency of 0.003% (1/31400) total alleles studied. The highest observed frequency was 0.118% (1/848) of Latino alleles. This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be deleterious by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
