The methionine at codon 173 is replaced by threonine, an amino acid with similar properties. An alternate amino acid substitution at this codon, p.M173V, has been reported in a pediatric onset hypertrophic cardiomyopathy case; however, clinical details were limited (Morita H et al. N. Engl. J. Med., 2008 May;358:1899-908). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
