This protein is an important cell cycle checkpoint kinase that phosphorylates; thus, it functions as a regulator of a wide variety of downstream proteins, including tumor suppressor proteins p53 and BRCA1, checkpoint kinase CHK2, checkpoint proteins RAD17 and RAD9, and DNA repair protein NBS1. This protein and the closely related kinase ATR are thought to be master controllers of cell cycle checkpoint signaling pathways that are required for cell response to DNA damage and for genome stability. This variant is denoted ATM c.8617G>A at the cDNA level, p.Val2873Ile (V2873I) at the protein level, and results in the change of a Valine to an Isoleucine (GTA>ATA). This variant has been observed in at least one individual with a personal and/or family history suggestive of a hereditary cancer syndrome undergoing genetic testing via a multi-gene panel and in an individual with melanoma (Mu 2016, Yehia 2018). ATM Val2873Ile was not observed at a significant allele frequency in large population cohorts (Lek 2016). This variant is located in the catalytic region of the Kinase domain (Lavin 2004, Stracker 2013). In silico analysis, which includes protein predictors and evolutionary conservation, supports that this variant does not alter protein structure/function. Based on currently available evidence, it is unclear whether ATM Val2873Ile is a pathogenic or benign variant. We consider it to be a variant of uncertain significance. -
