MUTYH (HGNC:7527): (mutY DNA glycosylase) This gene encodes a DNA glycosylase involved in oxidative DNA damage repair. The enzyme excises adenine bases from the DNA backbone at sites where adenine is inappropriately paired with guanine, cytosine, or 8-oxo-7,8-dihydroguanine, a major oxidatively damaged DNA lesion. The protein is localized to the nucleus and mitochondria. This gene product is thought to play a role in signaling apoptosis by the introduction of single-strand breaks following oxidative damage. Mutations in this gene result in heritable predisposition to colorectal cancer, termed MUTYH-associated polyposis (MAP). This variant disrupts the p.Gly286 amino acid residue in MUTYH. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 17703316, 18422726, 25820570, 25892863). The glycine at codon 286 is replaced by valine, an amino acid with dissimilar properties. Based on an internal structural analysis using a macromolecular structure from the RCSB Protein Data Bank, this variant is more disruptive than nearby pathogenic MUTYH variants (Burley SK et al. Nucleic Acids Res., 2019 Jan;47:D464-D474). This amino acid position is highly conserved in available vertebrate species. In addition, p.G286V is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
