The glycine at codon 286 is replaced by valine, an amino acid with dissimilar properties. Based on an internal structural analysis using a macromolecular structure from the RCSB Protein Data Bank, this variant is more disruptive than nearby pathogenic MUTYH variants (Burley SK et al. Nucleic Acids Res., 2019 Jan;47:D464-D474). This amino acid position is highly conserved in available vertebrate species. In addition, p.G286V is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
