The p.I52L variant (also known as c.154A>C), located in coding exon 2 of the RAD51C gene, results from an A to C substitution at nucleotide position 154. The isoleucine at codon 52 is replaced by leucine, an amino acid with highly similar properties. This variant was previously detected in 0/1388 hereditary breast and/or ovarian cancer families and 1/427 controls (Thompson et al. Hum. Mutat. 2012; 33: 95-9). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
This missense variant replaces isoleucine with leucine at codon 52 of the RAD51C protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with RAD51C-related disorders in the literature and has been observed in one individual in a control group (PMID: 21990120). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
