The encoded protein is involved in the ubiquitination and degradation of hypoxia-inducible-factor (HIF), which is a transcription factor that plays a central role in the regulation of gene expression by oxygen. In addition to oxygen-related gene expression, this protein plays a role in many other cellular processes including cilia formation, cytokine signaling, regulation of senescence, and formation of the extracellular matrix. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This alteration (also known as c.553+1G>A and IVS+1G>A) was reported in multiple individuals meeting clinical criteria for Von Hippel-Lindau syndrome (VHLS) (Dollfus, 2002; Palui, 2019; Ambry internal data) and multiple additional individuals with primary features of VHLS (Olschwang, 1998; Cybulski, 2004; Rao, 2010; Zhou, 2010). This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site and may result in the creation or strengthening of a novel splice donor site.
