The c.1802C>G (p.A601G) alteration is located in exon 9 (coding exon 9) of the DNAAF5 gene. This alteration results from a C to G substitution at nucleotide position 1802, causing the alanine (A) at amino acid position 601 to be replaced by a glycine (G). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
This sequence change replaces alanine with glycine at codon 601 of the DNAAF5 protein (p.Ala601Gly). The alanine residue is moderately conserved and there is a small physicochemical difference between alanine and glycine. This variant is present in population databases (rs745537484, ExAC 0.009%). This variant has not been reported in the literature in individuals with DNAAF5-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
