The homotetramer enzyme catalyzes the initial step of the mitochondrial fatty acid beta-oxidation pathway. Defects in this gene cause medium-chain acyl-CoA dehydrogenase deficiency, a disease characterized by hepatic dysfunction, fasting hypoglycemia, and encephalopathy, which can result in infantile death. The variant allele was found at a frequency of 1.2e-05 in 251430 control chromosomes (gnomAD). c.320T>C has been reported in the literature in individuals affected with Medium Chain Acyl-CoA Dehydrogenase Deficiency (examples: Arnold_2010, Smith_2010, Hsu_2008, McKinney_2004). These data indicate that the variant is likely to be associated with disease. The following publications have been ascertained in the context of this evaluation (PMID: 20036593, 20434380, 15171998, 18450854).
