Charcot-Marie-Tooth disease Uncertain:1
- -
Inborn genetic diseases Uncertain:1
The c.604C>G (p.Q202E) alteration is located in exon 9 (coding exon 8) of the LRSAM1 gene. This alteration results from a C to G substitution at nucleotide position 604, causing the glutamine (Q) at amino acid position 202 to be replaced by a glutamic acid (E). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Charcot-Marie-Tooth disease axonal type 2P Uncertain:1
This sequence change replaces glutamine, which is neutral and polar, with glutamic acid, which is acidic and polar, at codon 202 of the LRSAM1 protein (p.Gln202Glu). This variant is present in population databases (rs746274685, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with LRSAM1-related conditions. ClinVar contains an entry for this variant (Variation ID: 408270). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt LRSAM1 protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
