This variant is denoted POLD1 c.1762G>A at the cDNA level, p.Glu588Lys (E588K) at the protein level, and results in the change of a Glutamic Acid to a Lysine (GAG>AAG). This variant has not, to our knowledge, been published in the literature as a pathogenic or benign germline variant. POLD1 Glu588Lys was not observed at a significant allele frequency in large population cohorts (Lek 2016). This variant is located in the polymerase domain and the Pol II/Motif A (Preston 2010). In silico analysis, which includes protein predictors and evolutionary conservation, supports a deleterious effect. Based on currently available evidence, it is unclear whether POLD1 Glu588Lys is a pathogenic or benign variant. We consider it to be a variant of uncertain significance. -

The p.E588K variant (also known as c.1762G>A), located in coding exon 13 of the POLD1 gene, results from a G to A substitution at nucleotide position 1762. The glutamic acid at codon 588 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
