The frequency of this variant in the general population, 0.000055 (7/128378 chromosomes, http://gnomad.broadinstitute.org), is uninformative in assessment of its pathogenicity. In the published literature, the variant has been reported in a patient suspected to have type 2M VWD in the published literature (PMID: 22102198 (2011)). Variant summary: VWF c.4000C>T (p.Arg1334Trp) results in a non-conservative amino acid change located in the von Willebrand factor, type A (IPR002035) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2.8e-05 in 250476 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.4000C>T has been reported in the literature in individuals affected with Von Willebrand Disease without strong evidence of causality (Sanda_2015, Vangenechten_2022). These reports do not provide unequivocal conclusions about association of the variant with Von Willebrand Disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications has been ascertained in the context of this evaluation (PMID: 36299619). One submitter has cited a clinical-significance assessment for this variant to ClinVar after 2014 and classified it as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance. -
