The lysine at codon 2058 is replaced by glutamine, an amino acid with similar properties. Among a cohort of 103 patients with amyotrophic lateral sclerosis (ALS), this alteration was reported in one patient with familial ALS (Cooper-Knock J et al. Front Mol Neurosci, 2017 Nov;10:370). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
