The p.Y508C variant (also known as c.1523A>G), located in coding exon 4 of the BAG3 gene, results from an A to G substitution at nucleotide position 1523. The tyrosine at codon 508 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
