The leucine at codon 1935 is replaced by isoleucine, an amino acid with highly similar properties. A different alteration at this position, p.L1935P, was reported in 1/565 individuals with a clinical diagnosis of neurofibromatosis, and was found to segregate in 3/3 affected individuals in the proband's family (Sabbagh A et al. Hum. Mutat., 2013 Nov;34:1510-8). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
