This sequence change replaces threonine, which is neutral and polar, with isoleucine, which is neutral and non-polar, at codon 1462 of the POLE protein (p.Thr1462Ile). This variant is present in population databases (rs747856376, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with POLE-related conditions. ClinVar contains an entry for this variant (Variation ID: 574322). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt POLE protein function with a negative predictive value of 80%. The p.T1462I variant (also known as c.4385C>T), located in coding exon 34 of the POLE gene, results from a C to T substitution at nucleotide position 4385. The threonine at codon 1462 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
