This variant has been reported in an individual affected with ovarian cancer (PMID: 21964575). This variant has been identified in 1/251298 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. - |

The lysine at codon 209 is replaced by arginine, an amino acid with highly similar properties. This variant was reported in one out of 549 ovarian cancer cases from the Netherlands but was not observed in 436 Icelandic and 284 Spanish ovarian cancer cases or in 1446 Dutch and 2554 Icelandic controls (Rafnar T et al. Nat. Genet. 2011 Nov;43:1104-7). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
