Several computational tools predict a significant impact on normal splicing: Two predict the variant abolishes a 3' acceptor site. Two predict the variant weakens a 3' acceptor site. Four predict the variant creates a 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 8e-06 in 251362 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.47-6C>A has been reported in the literature in individuals affected with Deficiency Of Butyryl-CoA Dehydrogenase. However, a second causative variant was not identified (Adhikari_2020, van Maldegem_2006). These report(s) do not provide unequivocal conclusions about association of the variant with Deficiency Of Butyryl-CoA Dehydrogenase.
