The p.P2234A variant (also known as c.6700C>G), located in coding exon 15 of the APC gene, results from a C to G substitution at nucleotide position 6700. The proline at codon 2234 is replaced by alanine, an amino acid with highly similar properties. This variant was identified in a cohort of 681 ancestrally diverse, healthy subjects (Bodian DL et al. PLoS One, 2014 Apr;9:e94554). This amino acid position is highly conserved in available vertebrate species. In addition, in silico predictors for this gene do not accurately predict pathogenicity.
