Genes affected
USH1C (HGNC:12597): (USH1 protein network component harmonin) This gene encodes a scaffold protein that functions in the assembly of Usher protein complexes. The protein contains PDZ domains, a coiled-coil region with a bipartite nuclear localization signal and a PEST degradation sequence. Defects in this gene are the cause of Usher syndrome type 1C and non-syndromic sensorineural deafness autosomal recessive type 18. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Mar 2009]

Uncertain significance, criteria provided, single submitterclinical testingWomen's Health and Genetics/Laboratory Corporation of America, LabCorpSep 23, 2021Variant summary: USH1C c.266G>A (p.Arg89His) results in a non-conservative amino acid change in the encoded protein sequence. Three of four in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 251052 control chromosomes. c.266G>A has been reported in the literature as a compound heterozygous genotype in at-least one family with two affected individuals affected with recessive, prelingual, severe to profound non syndromic hearing loss (NSHL) (example, Ganapathy_2014). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Three clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All laboratories classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic. -

Uncertain significance, criteria provided, single submitterclinical testingInvitaeAug 23, 2022This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 89 of the USH1C protein (p.Arg89His). This variant is present in population databases (rs749647539, gnomAD 0.006%). This missense change has been observed in individual(s) with USH1C-related conditions (PMID: 24416283). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 553785). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
