The encoded protein responds to diverse cellular stresses to regulate expression of target genes, thereby inducing cell cycle arrest, apoptosis, senescence, DNA repair, or changes in metabolism. Mutations in this gene are associated with a variety of human cancers, including hereditary cancers such as Li-Fraumeni syndrome. Alternative splicing of this gene and the use of alternate promoters result in multiple transcript variants and isoforms. The variant was identified in control databases in 4 of 277226 chromosomes at a frequency of 0.000014 in the following populations: African in 2 of 24024 chromosomes (freq: 0.00008), Latino in 1 of 34420 chromosomes (freq. 0.00003), and European (Non-Finnish) in 1 of 126726 chromosomes (freq. 0.000008) increasing the likelihood that this may be a low frequency benign variant in certain populations of origin (Genome Aggregation Consortium Feb 27, 2017).
