Genes affected
CDC73 (HGNC:16783): (cell division cycle 73) This gene encodes a tumor suppressor that is involved in transcriptional and post-transcriptional control pathways. The protein is a component of the the PAF protein complex, which associates with the RNA polymerase II subunit POLR2A and with a histone methyltransferase complex. This protein appears to facilitate the association of 3' mRNA processing factors with actively-transcribed chromatin. Mutations in this gene have been linked to hyperparathyroidism-jaw tumor syndrome, familial isolated hyperparathyroidism, and parathyroid carcinoma. [provided by RefSeq, Jul 2009]

CDC73-related disorder Uncertain:1
Uncertain significance, criteria provided, single submitterclinical testingPreventionGenetics, part of Exact SciencesFeb 01, 2023The CDC73 c.803G>A variant is predicted to result in the amino acid substitution p.Arg268Gln. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.0026% of alleles in individuals of European (Non-Finnish) descent in gnomAD (http://gnomad.broadinstitute.org/variant/1-193117070-G-A), and has been classified as uncertain in the ClinVar database (https://www.ncbi.nlm.nih.gov/clinvar/variation/462767). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
Hyperparathyroidism 1 Uncertain:1
Uncertain significance, criteria provided, single submitterclinical testingBaylor GeneticsMar 07, 2024- -
Parathyroid carcinoma Uncertain:1
Uncertain significance, criteria provided, single submitterclinical testingInvitaeDec 04, 2023This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 268 of the CDC73 protein (p.Arg268Gln). This variant is present in population databases (rs750706815, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with CDC73-related conditions. ClinVar contains an entry for this variant (Variation ID: 462767). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt CDC73 protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
Hereditary cancer-predisposing syndrome Uncertain:1
Uncertain significance, criteria provided, single submitterclinical testingAmbry GeneticsNov 15, 2022The p.R268Q variant (also known as c.803G>A), located in coding exon 8 of the CDC73 gene, results from a G to A substitution at nucleotide position 803. The arginine at codon 268 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
