The p.R297Q variant (also known as c.890G>A), located in coding exon 4 of the GALNT12 gene, results from a G to A substitution at nucleotide position 890. The arginine at codon 297 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear. -

This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 297 of the GALNT12 protein (p.Arg297Gln). This variant is present in population databases (rs751386354, gnomAD 0.08%), and has an allele count higher than expected for a pathogenic variant.
