This variant has not been reported in the literature in individuals with SYNE2-related disease. This variant is present in population databases (rs752110658, ExAC 0.01%), and has an allele count higher than expected for a pathogenic variant (PMID: 28166811). This sequence change replaces arginine with threonine at codon 2860 of the SYNE2 protein (p.Arg2860Thr). The arginine residue is weakly conserved and there is a moderate physicochemical difference between arginine and threonine.
