Inborn genetic diseases Uncertain:1
The p.G309S variant (also known as c.925G>A), located in coding exon 7 of the RUNX1 gene, results from a G to A substitution at nucleotide position 925. The glycine at codon 309 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
Hereditary thrombocytopenia and hematologic cancer predisposition syndrome Uncertain:1

Hereditary thrombocytopenia and hematological cancer predisposition syndrome associated with RUNX1 Uncertain:1
This variant is present in population databases (rs752288830, gnomAD 0.007%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on RUNX1 protein function. ClinVar contains an entry for this variant (Variation ID: 573788). This variant has not been reported in the literature in individuals affected with RUNX1-related conditions. This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 309 of the RUNX1 protein (p.Gly309Ser). -
