The p.K136I variant (also known as c.407A>T), located in coding exon 5 of the POLE gene, results from an A to T substitution at nucleotide position 407. The lysine at codon 136 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is conserved.
