The p.E1178V variant (also known as c.3533A>T), located in coding exon 19 of the BRIP1 gene, results from an A to T substitution at nucleotide position 3533. The glutamic acid at codon 1178 is replaced by valine, an amino acid with dissimilar properties. This variant was reported in 1/60,466 breast cancer cases and in 2/53,461 controls (Dorling et al. N Engl J Med. 2021 02;384:428-439). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This missense variant replaces glutamic acid with valine at codon 1178 of the BRIP1 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual undergoing genetic testing for hereditary breast and ovarian cancer (PMID: 38136308). In an international breast cancer case-control meta-analysis, this variant was detected in 1/60466 cases and 2/53461 unaffected controls (PMID: 33471991). This variant has been identified in 1/250712 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

This sequence change replaces glutamic acid, which is acidic and polar, with valine, which is neutral and non-polar, at codon 1178 of the BRIP1 protein (p.Glu1178Val). This variant is present in population databases (rs752850661, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with BRIP1-related conditions.
