The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.2857G>A has been reported in the literature in an individual affected with breast cancer (Dorling_2021). This report does not provide unequivocal conclusions about association of the variant with Hereditary Nonpolyposis Colorectal Cancer. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. One ClinVar submitter (evaluation after 2014) cites the variant as likely benign reporting evidence of co-occurence with a mutation in another gene that clearly explains a proband's phenotype (SCV000273243.5), while three other ClinVar submitters (evaluation after 2014) cite it as uncertain significance.
