Inborn genetic diseases Uncertain:1
The p.Y838C variant (also known as c.2513A>G), located in coding exon 19 of the BUB1B gene, results from an A to G substitution at nucleotide position 2513. The tyrosine at codon 838 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear. -
Mosaic variegated aneuploidy syndrome 1 Uncertain:1
In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This sequence change replaces tyrosine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 838 of the BUB1B protein (p.Tyr838Cys). This variant is present in population databases (rs753916523, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with BUB1B-related conditions. ClinVar contains an entry for this variant (Variation ID: 465372). -
