Alternative splicing results in two transcript variants encoding different isoforms. Other alternatively spliced transcripts have been described, but the full length sequences have not been determined. Several of these transcripts are predicted to encode truncated products which lack the TM and may be secreted. This sequence change replaces serine, which is neutral and polar, with arginine, which is basic and polar, at codon 270 of the PKHD1 protein (p.Ser270Arg). This variant is present in population databases (rs754448530, gnomAD 0.003%). This missense change has been observed in individual(s) with autosomal recessive polycystic kidney disease and/or PKHD1-related conditions (PMID: 27225849; Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 551838).
