The encoded protein responds to diverse cellular stresses to regulate expression of target genes, thereby inducing cell cycle arrest, apoptosis, senescence, DNA repair, or changes in metabolism. Mutations in this gene are associated with a variety of human cancers, including hereditary cancers such as Li-Fraumeni syndrome. Alternative splicing of this gene and the use of alternate promoters result in multiple transcript variants and isoforms. Additional isoforms have also been shown to result from the use of alternate translation initiation codons from identical transcript variants (PMIDs: 12032546, 20937277). Therefore, this variant has been classified as Likely Pathogenic. No experimental studies have been performed to test the effects of this specific variant on TP53 protein function. However, this variant deletes two amino acid residues from the tetramerization domain (residues 323-356) and all of the C-terminal regulatory domain (residues 363-393) of the TP53 protein. The C-terminal region of TP53 protein has been shown to be important for regulation, nuclear localization, DNA-binding, and transactivation activity (PMID: 22178617, 25794615, 26205489, 20932800). This variant has not been reported in the literature in individuals with TP53-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change results in a premature translational stop signal in the TP53 gene (p.Gln354*). While this is not anticipated to result in nonsense mediated decay, it is expected to delete the last 39 amino acids of the TP53 protein. - |
