The p.A97V variant (also known as c.290C>T), located in coding exon 3 of the SDHD gene, results from a C to T substitution at nucleotide position 290. The alanine at codon 97 is replaced by valine, an amino acid with similar properties. This alteration has been identified in an individual with a personal history of pheochromocytoma; however, functional studies demonstrated normal expression, immunohistochemistry, SDH enzymatic assay, and succinate/fumarate concentration ratio in the tumor (Canu L et al. J Clin Endocrinol Metab, 2014 Jul;99:2321-6; Richter S et al. Genet Med, 2019 Mar;21:705-717). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
Hereditary pheochromocytoma-paraganglioma Uncertain:1
This missense variant replaces alanine with valine at codon 97 of the SDHD protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with pheochromocytoma PCC). However, the tumor in this patient showed SDH activity and succinate to fumarate ratio resembled that of a wild-type tumor (PMID: 24758185). This variant has been identified in 1/251488 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
