Genes affected
MYH2 (HGNC:7572): (myosin heavy chain 2) Myosins are actin-based motor proteins that function in the generation of mechanical force in eukaryotic cells. Muscle myosins are heterohexamers composed of 2 myosin heavy chains and 2 pairs of nonidentical myosin light chains. This gene encodes a member of the class II or conventional myosin heavy chains, and functions in skeletal muscle contraction. This gene is found in a cluster of myosin heavy chain genes on chromosome 17. A mutation in this gene results in inclusion body myopathy-3. Multiple alternatively spliced variants, encoding the same protein, have been identified. [provided by RefSeq, Sep 2009]

Uncertain significance, criteria provided, single submitterclinical testingInvitaeJul 17, 2023This variant is present in population databases (rs756953958, gnomAD 0.007%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. ClinVar contains an entry for this variant (Variation ID: 183667). This missense change has been observed in individual(s) with proximal myopathy with ophthalmoplegia (PMID: 24193343). This sequence change replaces threonine, which is neutral and polar, with isoleucine, which is neutral and non-polar, at codon 178 of the MYH2 protein (p.Thr178Ile). -

Uncertain significance, criteria provided, single submitterclinical testingPreventionGenetics, part of Exact SciencesDec 12, 2023The MYH2 c.533C>T variant is predicted to result in the amino acid substitution p.Thr178Ile. This variant has been reported in the homozygous state in an individual with autosomal recessive myosin myopathy with external ophthalmoplegia (Tajsharghi. 2014. PubMed ID: 24193343). The patient’s parents were heterozygous for the variant and were reported to be unaffected. This variant is reported in 0.0062% of alleles in individuals of African descent in gnomAD. At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
