NKX2-5 (HGNC:2488): (NK2 homeobox 5) This gene encodes a homeobox-containing transcription factor. This transcription factor functions in heart formation and development. Mutations in this gene cause atrial septal defect with atrioventricular conduction defect, and also tetralogy of Fallot, which are both heart malformation diseases. Mutations in this gene can also cause congenital hypothyroidism non-goitrous type 5, a non-autoimmune condition. Alternative splicing results in multiple transcript variants. This variant results from an in-frame AAC deletion at nucleotide positions 871 to 873. This results in the in-frame deletion of an asparagine at codon 291. This variant was reported in one individual from a congenital heart defect cohort, as well as in an unaffected parent (McElhinney DB et al. J Am Coll Cardiol, 2003 Nov;42:1650-5). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be neutral by in silico analysis (Choi Y et al., PLoS ONE 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
