The p.Asp1502Gly variant in MYO7A has been previously reported in 1 individual w ith Usher syndrome; however, the individual was also reported to have 2 addition al truncating variants in MYO7A identified (Le Quesne Stabej 2012; https://grena da.lumc.nl/LOVD2/Usher_montpellier). This variant has been identified in 2/30782 South Asian chromosomes and in 1/18866 East Asian chromosomes by the Genome Agg regation Database (gnomAD, http://gnomad.broadinstitute.org; dbSNP rs757460257); however, its frequency is not high enough to rule out a pathogenic role. Comput ational prediction tools suggest the possible creation of a cryptic 5' splice si te; however, this information is not predictive enough to determine pathogenicit y. Additional computational prediction tools and conservation analysis suggest that this variant may impact the protein, though this information is not predict ive enough to determine pathogenicity. In summary, the clinical significance of the p.Asp1502Gly variant is uncertain. -
Variant summary: MYO7A c.4505A>G (p.Asp1502Gly) results in a non-conservative amino acid change located in the FERM domain (IPR000299) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 249006 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.4505A>G has been reported in the literature in cis with an upstream loss of function nonsense variant, c.1258A>T (p.Lys420*) in a compound heterozygous genotype with a different MYO7A variant in two unrelated families affected with hearing loss (example, Richards_2019, Doll_2020). These report(s) do not provide unequivocal conclusions about association of the variant with Usher Syndrome. At-least two co-occurrences in cis with another pathogenic variant(s) have been reported (MYO7A c.1258A>T, p.Lys420* as summarized above), providing supporting evidence for a benign role. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
